Drug intelligence / Profile preview
[68Ga]Ga-NOTA-5F7 is an investigational radiolabeled single-domain antibody fragment (nanobody) designed for positron emission tomography (PET) imaging of human epidermal growth factor receptor 2 (HER2) expression. It consists of the anti-HER2 camelid-derived nanobody 5F7 conjugated to the chelator 1,4,7-triazonane-1,4,7-triacetic acid (NOTA) and labeled with the positron-emitting radionuclide gallium-68 (68Ga). Originally developed at Duke University and clinically translated by Fudan University and Guizhou Medical University, the tracer is being evaluated for non-invasive, whole-body quantification of HER2 expression in patients with HER2-positive breast cancer and metastatic urothelial carcinoma. It demonstrates high sensitivity and potential for detecting intracranial metastases, and is paired with the therapeutic/SPECT agent [131I]SGMIB-5F7 as part of a targeted theranostic strategy.
